ABSTRACT
RNA interference (RNAi) by introducing double-stranded (dsRNA) is a powerful approach to the analysis of gene function in insects; however, RNAi responses vary dramatically different insect species and tissues, underlying mechanisms remain poorly understood. The migratory locust, destructive pest hemimetabolic with panoistic ovaries, considered be highly susceptible via dsRNA injection, but its ovary appears completely insensitive. In present study, we showed that persisted only briefly locust haemolymph. ovariole sheath was permeable dsRNA, injected not follicle cells oocytes. lack uptake into oocytes likely primary factor contributes ineffective response ovaries. These observations provide insights tissue-dependent variability help achieving successful silencing insensitive tissues.
