ABSTRACT
Summary The maize ( Zea mays L.) rum1‐R rootless with undetectable meristems 1‐Reference ) mutant does not initiate embryonic seminal roots and post‐embryonic lateral at the primary root. Map‐based cloning revealed that Rum1 encodes a 269 amino acid (aa) monocot‐specific Aux/IAA protein. protein lacks 26 acids including GWPPV degron sequence in domain II part of bipartite NLS (nuclear localization sequence). Significantly reduced root density (approximately 35%) heterozygous plants suggests is semi‐dominant mutant. Overexpression under control MSY M ethionine SY nthase) promoter supports this notion by displaying number (31–37%). Functional characterization auxin‐inducible localizes to nucleus. Moreover, RUM1 unstable half life time approximately 22 min while very stable. In vitro vivo experiments demonstrated an interaction ZmARF25 ZmARF34 Z. AUXIN RESPONSE FACTOR 25 34). summary, presented data suggest canonical required for initiation maize.
