ABSTRACT
The influence of 12( S )‐hydroxyicosatetraenoic acid (12‐HETE), that we have previously shown to decrease the proliferative response human lymphocytes mitogens, on diacylglycerol and phosphatidic (PtdOH) formation was investigated in stimulated peripheral blood mononuclear cells (PBMC). When PBMC were first enriched with 12‐HETE, then by mitogenic lectin concanavalin A (Con A), production PtdOH normally associated Con stimulation markedly increased as compared non‐enriched cells. Con‐A‐induced rise mass decreased 1 % ethanol 12‐HETE‐enriched cells, whereas it unaffected control alone. Furthermore, [ 3 H]arachidonic‐acid‐labelled phosphatidylalcohol significantly upon stimulation, no being synthesized Collectively, these results suggest that, presence stimulates a phospholipase D activity which not triggered These data are consistent lack effect suramin, reported inhibitor, observed alone suramin‐induced 12‐HETE‐plus‐Con‐A‐treated Moreover, 12–[ H]HETE‐enriched produced significant amount H]HETE‐containing (0.4% total PtdOH) resting conditions. Upon Stimulation A, phorbol ester tetrade‐canoylphorbol acetate or anti‐CD3 mAb OKT3, this proportion 0.1–0.2%, since more drastically than 12‐HETE‐containing species. Although present relatively low species might been generated strategic compartments membrane bilayer so following events involved transduction signal could be impaired. GC analyses pointed out drastic variations fatty composition Especially contained higher saturated acids lower arachidonic formed Such content very likely potential.
