ABSTRACT
The present study was conducted by screening zein-degrading bacteria in an attempt to obtain protease.Soil were screened formation of a clear zone on zein plates. Characterization bacterium indicated taxonomic affiliation Bacillus pumilus, and named MS-1 strain. strain produced two different types extracellular proteases, BPP-A BPP-B. In this study, we purified characterized because it exhibited higher ability hydrolyze than When casein used as the substrate, optimal pH for 11.0. BPP-A, better substrate at 13.0, whereas one bppA gene encoded 383-amino acid pre-pro form mature contained 275 amino residues. It concluded that belonged subtilisin family.A assigned B. pumilus extreme alkaline condition. This is first report micro-organisms. subtilisin-like protease possible utilize industrial enzyme production hydrolysates.
