ABSTRACT
To design and validate a colorimetric loop‐mediated isothermal amplification assay for rapid detection of Phytophthora infestans DNA. Two sets (LAMP) primers were designed evaluated their sensitivity specificity P. infestans. ITSII targeted portion the internal transcribed spacer region ribosomal These had limit 2 pg DNA cross‐reacted with closely related species nicotianae. Rgn86_2 primers, to improve specificity, conserved hypothetical protein. 200 did not cross‐react The was tested further using andina, ipomoeae, mirabilis phaseoli. Cross‐reactions occurred andina mirabilis, but neither occurs on tomato or potato. Both primer able detect extracted from late blight leaf lesions. LAMP assays detected pure cultures as well infected tissue. higher sensitivity, specificity. This is first report infestans, causal organism potato blight. have potential immediate utility in plant disease research diagnostic laboratories.
