ABSTRACT
Emerging evidence have illustrated the vital roles of long noncoding RNAs (lncRNAs) in glioma. Nevertheless, majority their and mechanisms gliomagenesis are still largely unclear. In this study, we investigate lncRNA CASC9 on glioma tumourigenesis authenticate its potential mechanisms. Results manifested that was highly expressed specimens cells, moreover, ectopic overexpression correlated with patients' clinic. Functional studies found siRNA-mediated silencing inhibited proliferative ability, invasion vitro, impaired tumour growth vivo. Mechanical revealed miR-519d both targeted 3'-UTR STAT3 mRNA, which identified by luciferase reporter assay RNA immunoprecipitation (RIP). Moreover, chromatin (ChIP) STAT3, an oncogenic transcription factor, could bind promoter activate transcriptional level. conclusion, our results concluded promotes expression via sponging miR-519d, return, transcription, forming a positive feedback loop CASC9/miR-519d/STAT3.
