ABSTRACT
Proteasome 26S subunit ATPase 4 (PSMC4) could regulate cancer progression. However, the function of PSMC4 in prostate carcinoma (PCa) progression requires further clarification. In study, and chromobox 3 (CBX3) levels were verified by TCGA data tissue microarrays. Cell counting kit-8, cell apoptosis, cycle, wound healing, transwell xenograft tumour model assays performed to verify biological functions PCa. RNA-seq, PCR, western blotting co-IP mechanism PSMC4. Results showed that level was significantly increased PCa tissues, patients with a high exhibited shorter overall survival. knockdown markedly inhibited proliferation, cycle migration vitro vivo, promoted apoptosis. Then study revealed CBX3 downstream target reduced level, PI3K-AKT-mTOR signalling. overexpression epidermal growth factor receptor (EGFR) level. Finally, reverse effect DU145 cells, effects on clonal formation rescued knockdown, regulated EGFR-PI3K-AKT-mTOR conclusion, mediating CBX3-EGFR-PI3K-AKT-mTOR pathway. These findings provided new for treatment.
