ABSTRACT
The modulation of the kisspeptin system holds promise as a treatment for human reproductive disorders and managing livestock breeding. design analogs has overcome some unfavorable properties endogenous ligands. However, applications requiring prolongation drug activity, such ovulation induction in ewe during non-breeding season, additional improvement is required. To this aim, we designed tested three formulations containing analog C6. Two were based on polymeric nanoparticles (NP1 NP2) third was hydrogels composed mixture cyclodextrin polymers dextran grafted with alkyl side chains (MD/pCD). Only MD/pCD formulation prolonged C6 shown by monitoring luteinizing hormone (LH) plasma concentration (elevation duration 23.4 ± 6.1, 13.7 4.7 12.0 2.4 h MD/pCD, NP1 NP2, respectively). When compared free (15 nmol/ewe), formulated (MD/pCD) doses 10, 15 30 nmol/ewe, but not 90 nmol/ewe dose, provided more gradual release an attenuated LH first 6 post-treatment. season without progestogen priming, only, dose triggered (50% ewes). Hence, showed that adapted action time would improve efficacy respect to inducing season. This result suggests might find management reproduction also point possibility their use pathologies.
