ABSTRACT
The role of Ras guanine nucleotide-releasing protein 1 (RasGRP1) in tumourigenesis has been a subject debate, and its functions clinical significance hepatocellular carcinoma (HCC) remain unknown. Here, we evaluated the expression RasGRP1 HCC determined how it contributes to cell proliferation.RasGRP1 was measured by quantitative polymerase chain reaction (qPCR) Western blotting 24 paired tissues para-tumour tissues. confirmed immunohistochemical analysis tissue microarray from independent cohort. Overall survival (OS) disease-free (DFS) were estimated using Kaplan-Meier method, risk factors that contributed OS or DFS identified Cox regression analysis. biologic relevance examined small interfering RNAs an exogenous plasmid construct. Chromatin immunoprecipitation assays performed examine binding Sp1 promoter.Increased associated with tumour size (P = .004), tumour-node-metastasis stage .032), Barcelona Clinic Liver Cancer .002). overexpression prognostic factor patients. downregulation inhibited proliferation, whereas promoted proliferation. Moreover, specificity bound promoter transcription. In addition, enhanced activation c-Raf pathway.RasGRP1 is upregulated promotes Thus, may be novel therapeutic target for HCC.
