ABSTRACT
Summary Plant establishment requires the formation and development of an extensive root system with architecture modulated by complex genetic networks. Here, we report identification PtrXB38 gene as expression quantitative trait loci (eQTL) hotspot, mapped using 390 leaf 444 xylem Populus trichocarpa transcriptomes. Among predicted targets this trans ‐eQTL were genes involved in plant hormone responses development. Overexpression led to significant increases callusing both stem‐born roots base‐born adventitious roots. Omics studies revealed that proteins controlling auxin transport signaling PtrXB38‐mediated formation. Protein–protein interaction assays indicated interacts components endosomal sorting complexes required for machinery, implying PtrXB38‐regulated may be mediated regulating endocytosis pathway. Taken together, work identified a crucial regulator sheds light on discovery other developmental regulators through combining eQTL mapping omics approaches.
