ABSTRACT
This study aimed to explore the effects of metformin on osteogenic differentiation alveolar bone marrow mesenchymal stem cells (BMSCs) from type-2 diabetes mellitus (T2DM) patients (DM-BMSCs) and implant osseointegration in rats, screen optimal concentration, investigate whether could protect against adverse impact T2DM BMSC capacity.Different concentrations were administered human-derived BMSCs Wistar rats receiving implants. ALP detection, alizarin red staining, real-time RT-PCR Western blotting performed detect osteogenesis mechanism. Toluidine blue staining was analyse bone-implant contact rats.Metformin increased a rat model promoted capacity DM-BMSCs via AMPK/BMP/Smad signalling pathway, 125 μM concentration; however, over 200 µM, showed an inhibitory effect DM-BMSCs. Additionally, at concentration (125 µM) identified this compensate for negative impacts BMSCs.Metformin can promote it might be effective drug increasing success rate T2DM-associated
