ABSTRACT
D1-cytoplasmic male sterility (CMS) rice is a sporophytic cytoplasmic male-sterile developed from Dongxiang wild that exhibits no-pollen-grain phenotype. A mitochondrial chimeric gene (orf182) was detected by genome sequencing and comparative analysis. Orf182 composed of three recombinant fragments, the largest which homologous to Sorghum bicolor sequences. In addition, orf182 found only in species collected China. Northern blot analysis showed transcripts were affected Rf genes isocytoplasmic restorer line DR7. Western ORF182 product localized mitochondria CMS line. An expression cassette containing fused transit peptide induced maintainer sterility, lacked pollen grains anthers. Furthermore, vivo also inhibited growth Escherichia coli, with lower respiration rate, excess accumulation reactive oxygen decreased ATP levels. We conclude possesses unique structure origin differing other identified genes, this connected non-pollen type D1-CMS rice.
