ABSTRACT
Historically, red cell concentrates (RCCs) have been manually glycerolized and deglycerolized using an open system (COBE 2991, Terumo). Implementation of a closed processor (ACP-215, Haemonetics) for glycerolization deglycerolization RCCs creates challenge management the historic cryopreserved RCC inventory. A study was undertaken to determine whether frozen could be processor, as processors are being discontinued. Thirteen ABO/Rh matched were pooled split produce six large (approximately 354 mL) small 244 RCCs. All units stored 14 days post-collection, at ≤ -65°C ≥72 h. Half each size COBE 2991 resuspended in 0.9% saline, remaining centrifuged, on ACP-215, AS-3. RBC quality tested 24 ± 2 h post-deglycerolization. ACP-215 had significantly lower hemolysis (p < .001) levels than those processed COBE2991. Large hematocrits .05), hemoglobin .01), recovery = did 2991. met regulatory standards hemolysis, hematocrit, hemoglobin, recovery. The closed-system reduced post-deglycerolization all units, content units. combination with centrifugation step, is suitable processing that glycerolized.
