ABSTRACT
1. Interlobular ducts were isolated from the rat pancreas and maintained in short‐term tissue culture. Fluid secretion these was measured using micropuncture techniques, intracellular calcium concentration ([Ca2+]i) by fura‐2 microspectrofluorimetry, cyclic AMP radioimmunoassay. 2. Applying secretin ACh simultaneously to caused either a stimulation or an inhibition of fluid depending on doses employed. 3. The inhibitory effect could be relieved atropine, protein kinase C (PKC) inhibitors staurosporine 1‐(5‐isoquinolinylsulphonyl)‐2‐methyl‐piperazine (H‐7). 4. Activation PKC 12‐O‐tetradecanoylphorbol‐13‐acetate (TPA) phorbol 12, 13‐dibutyrate (PDBu) inhibited secretin‐evoked secretion. 5. TPA also stimulated adenylate cyclase activator, forskolin. 6. Neither nor activators had any increase [Ca2+]i evoked 7. We conclude that combined ductal is probably mediated at point secretory mechanism distal generation messengers.
