High-throughput label-free molecular fingerprinting flow cytometry

ABSTRACT
Flow cytometry is an indispensable tool in biology for counting and analyzing single cells large heterogeneous populations. However, it predominantly relies on fluorescent labeling to differentiate and, hence, comes with several fundamental drawbacks. Here, we present a high-throughput Raman flow cytometer microfluidic chip that chemically probes live label-free manner. It based rapid-scan Fourier-transform coherent anti-Stokes scattering spectrometer as optical interrogator, enabling us obtain the broadband molecular vibrational spectrum of every cell fingerprint region (400 1600 cm-1) record-high throughput ~2000 events/s. As practical application method not feasible conventional cytometry, demonstrate single-cell analysis astaxanthin productivity photosynthetic dynamics Haematococcus lacustris.
