ABSTRACT
CAR T cell therapy has revolutionized the treatment of a spectrum blood-related malignancies. However, responses vary among cancer types and patients. Accurate monitoring dynamics is crucial for understanding evaluating efficacy. Positron emission tomography (PET) offers comprehensive view homing, especially in critical organs such as lymphoid structures bone marrow. This information will help assess response predict relapse risk. Current PET imaging methods require genetic modifications, limiting clinical use. To overcome this, we developed an antigen-based approach enabling whole-body imaging. The probe detects cells vivo without affecting their function. In immunocompetent B leukemia model, CAR-PET signal spleen predicted early mortality allows assessment altering established protocols. It seamlessly integrates with FDA-approved future generations, facilitating broader application.
