ABSTRACT
Postoperative rehemorrhage following intracerebral hemorrhage surgery is intricately associated with a high mortality rate, yet there now no effective clinical treatment. In this study, we developed hemoglobin (Hb)-responsive in situ implantable DNA hydrogel comprising Hb aptamers cross-linked two complementary chains and encapsulating deferoxamine mesylate (DFO). Functionally, the generates signals upon postoperative by capturing Hb, demonstrating distinctive "self-diagnosis" capability. addition, ongoing capture of mediates gradual disintegration hydrogel, enabling on-demand release DFO without compromising physiological iron-dependent functions. This process achieves self-treatment inhibiting ferroptosis neurocytes. collagenase autologous blood injection model-induced mimic model, exhibited 5.58-fold increase iron absorption efficiency, reducing hematoma size significantly (from 8.674 to 4.768 cubic millimeters). innovative Hb-responsive not only offers therapeutic intervention for but also provides self-diagnosis feedback, holding notable promise enhancing outcomes.
