ABSTRACT
Viral RNA synthesis of mononegaviruses occurs in cytoplasmic membraneless organelles called inclusion bodies (IBs). Here, we report that IBs mumps virus (MuV), which is the causative agent and belongs to family Paramyxoviridae , displayed liquid organelle properties formed by liquid-liquid phase separation. Super-resolution microscopic analysis MuV demonstrated nucleocapsid phospho (P)-proteins a cage-like structure viral polymerase adopted reticular pattern colocalized with RNAs. In addition, characterized host RNAs localized spatial transcriptome analysis, found containing G-quadruplex motif sequences (G4-RNAs) were concentrated. An vitro separation assay showed G4-RNAs interacted P protein enhanced condensation droplets. Together, our data show generates characteristic play an important role forming IBs.
