ABSTRACT
Despite remarkable recent achievements of immunotherapy strategies in cancer treatment, clinical responses remain limited to subsets patients. Predictive markers disease course and response are urgently needed. Recent results have revealed the potential predictive value immune cell phenotype spatial distribution at tumor site, prompting need for multidimensional immunohistochemical analyses tissues. To address this need, we developed a sample-sparing, highly multiplexed immunohistochemistry technique based on iterative cycles tagging, image scanning, destaining chromogenic substrate single slide. This assay, combination with newly automated digital landscaping solution, democratizes access high-dimensional by capturing complexity immunome using routine pathology standards. Applications method extend beyond screen validate comprehensive panels tissue-based prognostic markers, perform in-depth situ monitoring therapies, identify targets disease.
