Isolation of Bifidobacteria from Breast Milk and Assessment of the Bifidobacterial Population by PCR-Denaturing Gradient Gel Electrophoresis and Quantitative Real-Time PCR

ABSTRACT
The objective of this work was to elucidate if breast milk contains bifidobacteria and whether they can be transmitted the infant gut through breastfeeding. Twenty-three women their respective infants provided samples feces, respectively, at days 4 7 after birth. Gram-positive catalase-negative isolates from specific media with typical bifidobacterial shapes were identified genus level by F6PPK (fructose-6-phosphate phosphoketolase) assays species 16S rRNA gene sequencing. Bifidobacterial communities in assessed PCR-denaturing gradient gel electrophoresis (PCR-DGGE), levels estimated quantitative real-time PCR (qRTi-PCR). Bifidobacteria present 8 21 fecal samples. Bifidobacterium breve, B. adolescentis, bifidum isolated samples, while feces also contained longum pseudocatenulatum. PCR-DGGE revealed presence one four dominant bands 22 Sequences similarities above 98% as longum, bifidum, dentium. DNA detected qRTi-PCR same a range between 40 10,000 copies per ml. In conclusion, human seems source living for gut.
