ABSTRACT
MalF has been shown to be required for virulence in the important avian pathogen Mycoplasma gallisepticum To characterize function of MalF, predicted part a putative ABC transporter, we compared metabolite profiles mutant with transposon inserted malF (MalF-deficient ST 04-1; ΔmalF) those wild-type bacteria using gas chromatography-mass spectrometry and liquid spectrometry. Of substrates likely transported by an transport system, glycerol was detected at significantly lower abundance ΔmalF mutant, wild type. Stable isotope labeling [U-13C]glycerol reverse transcription-quantitative PCR analysis indicated that responsible import into M. that, absence transcription gtsA, which encodes second GtsA, upregulated, potentially increase glycerol-3-phosphate cell compensate loss MalF. The appeared have global effect on metabolism, suggesting it may also play regulatory role, cellular morphology affected, indicating change metabolism broader organization. Overall, this study suggests reduced is due perturbed uptake operon including should reannotated as golABC reflect its transport.IMPORTANCE Many mycoplasmas are pathogenic cause disease humans animals. causes chronic respiratory chickens infectious sinusitis turkeys, resulting economic losses poultry industries throughout world. Expanding our knowledge about pathogenesis mycoplasma infections requires better understanding specific gene functions these bacteria. In study, characterized metabolic protein involved pathogenicity gallisepticum, well expression selected genes, phenotype, H2O2 production. This key step forward elucidating why plays role chickens. emphasizes importance functional characterization proteins, tools such metabolomics, since prediction based homology other bacterial proteins not always accurate.
