ABSTRACT
ABSTRACT Pneumolysin (PLY) is an important virulence factor of Streptococcus pneumoniae . We examined the ability three murine monoclonal antibodies (MAbs) to PLY (PLY-4, PLY-5, and PLY-7) affect course pneumococcal pneumonia in mice. The intravenous administration PLY-4 PLY-7 protected mice from lethal effect purified toxin. Mice treated with before intranasal inoculation S. type 2 survived longer (median survival time, 100 h) than did untreated animals 60 ( P < 0.0001). median time for a combination was 130 h, significantly that given isotype-matched indifferent MAbs = 0.0288) or nontreated 0.0002). (>480 PLY-5 (48 h; 0.0001), (78 0.0007), (100 0.0443) alone. Similarly, rate (10 20 mice) higher obtained (1 20; 0.0033), (2 0.0138), (3 0.0407) These results suggest anti-PLY act synergistic effect. Furthermore, MAb associated significant decrease bacterial lung colonization lower frequencies bacteremia tissue injury respect control groups.
