ABSTRACT
ABSTRACT Infections with RNA viruses are sensed by the innate immune system through membrane-bound Toll-like receptors or cytoplasmic helicases RIG-I and MDA-5. It is believed that MDA-5 crucial for sensing infections picornaviruses, but there have been no studies on role of this protein during infection poliovirus, prototypic picornavirus. Beginning at 4 h postinfection, degraded in poliovirus-infected cells. Levels declined beginning 6 after rhinovirus type 1a encephalomyocarditis virus, was stable cells infected 16 echovirus 1. Cleavage not carried out either poliovirus proteinase 2A pro 3C . Instead, degradation occurs a proteasome- caspase-dependent manner. Degradation correlates cleavage poly(ADP) ribose polymerase (PARP), hallmark apoptosis. Induction apoptosis puromycin leads to both PARP The product observed treated ∼90 kDa, similar size putative Poliovirus-induced may be mechanism antagonize production I interferon response viral infection.
