ABSTRACT
Rheumatoid arthritis (RA) is a chronic autoimmune disease that causes joint deformity and disability. Deer velvet antler (DA), traditional Chinese medicine, has been used to treat various types of for several thousands years, but the underlying mechanisms are unknown. Herein, we investigated anti-arthritic anti-inflammatory effects DA in vitro vivo. The ethyl acetate layer ethanol extract (DA-EE-EA) was tumor necrosis factor (TNF)-[Formula: see text]-stimulated fibroblast-like synoviocyte MH7A cells, collagen-induced DBA/1 mice, SKG mice with zymosan-induced arthritis. DA-EE-EA reduced nitric oxide production, prostaglandin E2 levels, levels pro-inflammatory cytokines including interleukin (IL)-1[Formula: text], IL-6, IL-8 cells. also downregulated phosphorylation mitogen-activated protein kinase p38 c-Jun N-terminal translocation nuclear kappa B p65. Intraperitoneal injection 3 weeks substantially clinical scores vivo models. Pathohistological images hind paws showed immune cell infiltration, synovial hyperplasia, cartilage damage. cytokines, such as alpha, IL-1[Formula: IL-8, IL-17A, interferon-gamma, decreased paw homogenates DA-EE-EA-treated mice. We identified potential components, hexadecanamide, oleamide, erucamide, lysophosphatidylcholines, might contribute DA-EE-EA. In conclusion, RA by regulating inflammatory responses. However, individual components need further investigation future studies.
