ABSTRACT
VEGF is a highly specific stimulator of endothelial cells and may play an important role in angiogenesis the process tissue regeneration. We previously showed that cyclooxygenase-2 (COX-2) expressed mesenchymal ulcer bed involved repair process. To clarify COX-2 during gastric healing, we investigated relation between expression production human fibroblasts vivo vitro. Gastric were cultured RPMI 1640 with without IL-1alpha or IL-1beta presence absence NS-398, selective inhibitor. Supernatant PGE(2) concentrations measured by enzyme-linked immunosorbent assay. was determined Western blot analysis. surgical resections examined immunohistochemically. IL-1 dose dependently enhanced release after 24-h stimulation. also stimulated via induction. NS-398 significantly suppressed from IL-1-stimulated fibroblasts; concurrent addition restored NS-398-inhibited release. immunoreactivity colocalized fibroblast-like tissues. These results suggest plays key vitro induced COX-2-VEGF pathway might be healing.
