ABSTRACT
This study aimed to clarify epigenetic and genetic alterations that occur during renal carcinogenesis. The original method includes chromosome 3 specific NotI-microarrays containing 180 NotI-clones associated with 188 genes for hybridization 23 paired normal/tumor DNA samples of primary clear cell carcinomas (ccRCC). Twenty-two showed methylation and/or deletion in 17–57% tumors. These include tumor suppressors or candidates ( VHL, CTDSPL, LRRC3B, ALDH1L1 , EPHB1 ) were not previously considered as cancer-associated (e.g., LRRN1, GORASP1, FGD5 PLCL2 ). Bisulfite sequencing analysis confirmed a frequent event ccRCC. A set six markers NKIRAS1/RPL15, GORASP1/TTC21A VHL was suggested ccRCC detection biopsies. mRNA level decrease shown 6 NotI-associated using quantitative PCR: FOXP1, FGD5, PLCL2, . majority examined distinct expression profiles papillary RCC. strongest extent frequency downregulation gene both Moreover, the more pronounced histological types RCC stage III compared stages I II (P=0.03). same observed id="M2">P<0.06).
