Variety of RNAs in Peripheral Blood Cells, Plasma, and Plasma Fractions

Human peripheral blood contains RNA in cells and extracellular membrane vesicles, microvesicles exosomes, as well cell-free ribonucleoproteins. Circulating mRNAs noncoding RNAs, being internalized, possess the ability to modulate vital processes recipient cells. In this study, with SOLiD sequencing technology, we performed identification, classification, quantification of RNAs from fractions: cells, plasma, plasma vesicles pelleted at 16,000<mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML" id="M1"><mml:mrow><mml:mi>g</mml:mi></mml:mrow></mml:math>and 160,000<mml:math id="M2"><mml:mrow><mml:mi>g</mml:mi></mml:mrow></mml:math>, vesicle-depleted supernatant healthy donors non-small cell lung cancer (NSCLC) patients. It was determined that id="M3"><mml:mrow><mml:mi>g</mml:mi></mml:mrow></mml:math>blood were enriched mitochondria a set mitochondrial RNAs. The variable id="M4"><mml:mrow><mml:mi>g</mml:mi></mml:mrow></mml:math>pellets reflected prominent contribution U1, U5, U6 small nuclear RNAs’ fragments same time characterized by remarkable depletion nucleolar Besides microRNAs, variety snoRNAs dominated circulating differentially expressed fractions NSCLC Taken together, our data emphasize not only microRNAs but also messenger nuclear/nucleolar represent classes regulatory ncRNAs promising biomarkers for development disease diagnostic approaches.
