ABSTRACT
Hidradenitis suppurativa (HS) is a chronic inflammatory skin disease. Patients can present with nodules, abscesses up to fistulas, or sinus tracts in intertriginous body parts. Occlusion of the sebaceous gland unit leads its rupture, subsequent exuberant immune response. Given there still no causative therapy, better understand HS and develop novel therapeutic concepts, research activities field are constantly growing. Primary cells, blood ex vivo explant cultures from patients have been previously used as cell culture models. In vitro reconstituted epidermal models established study dermatoses, such psoriasis atopic dermatitis. For HS, exploration epidermis would be an excellent addition, e.g., biomarkers barrier function testing new topic treatment options. We therefore stratified model based on primary cells lesions. After isolating keratinocytes lesional skin, we cultured them submerged transwell system. To induce differentiation, then lifted air-liquid interface. Immunohistochemical staining demonstrated that our HS-epidermis meets expected differentiation pattern. detected secretion cytokines interleukin-1β TNF-α.
