<div>AbstractPurpose:<p>The Cancer Immune Monitoring and Analysis Centers – Immunologic Data Commons (CIMAC-CIDC) Network is supported by the NCI to identify biomarkers of response cancer immunotherapies across clinical trials using state-of-the-art assays. A primary platform for CIMAC-CIDC studies cytometry time flight (CyTOF), performed at all CIMAC laboratories. To ensure ability generate comparable CyTOF data labs, a multistep cross-site harmonization effort was undertaken.</p>Experimental Design:<p>We first harmonized standard operating procedures (SOPs) sites. Because new acquisition protocol comparing original narrow- or wide-bore injector introduced vendor (Fluidigm), we also tested this sites before finalizing SOP. We then assay experiments five shared cryopreserved one lyophilized internal control peripheral blood mononuclear cell (PBMC) with antibody cocktail consisting 14 isotype-tagged antibodies previously validated, plus additional liquid antibodies. These reagents samples were distributed centrally analyzed manual gating automated methods (Astrolabe).</p>Results:<p>Average coefficients variation (CV) each population reported compared previous multisite study. reached an intersite CV under 20% most subsets, very similar published study.</p>Conclusions:<p>These results establish reproduce in multicenter trials, highlight importance quality procedures, such as use spike-in samples, tracking variability assay.</p></div>
