ABSTRACT

Sigma1 KD suppresses DHT induced prostate cancer cell proliferation and tumor growth despite (or due to) increased autophagy. A,In vitro assay of Sigma1 shRNA-transduced LNCaP C4-2 cells precultured in CSS medium for 3 days then treated 6 with 1 nmol/L DHT. Live were counted by Trypan blue exclusion at the start time course (day 0), treatment. Data are represented as fold induction over control shRNA day 0. Datapoints represent mean increase number from least three independent determinations, error bars SEM. Two distinct clones, #4 #5, tested produced similar results. B, Immunoblots whole-cell protein extracts parallel culture performed using same experimental treatment conditions A. shown clone #5 cultures. C, Immunoblot transduced immediately prior to subcutaneous flank implantation into SCID mice. D, infected (#4, #5) (#1) injected right left flanks Tumor volume was measured caliper 12 weeks after implantation, sacrificing six tumors each condition, E, weight postinjection harvest. *, P < 0.05; **, 0.01. F, Oil Red O staining (clone #1) xenografted tumors.
