ABSTRACT

Abstract
Fatty acid synthase (FASN) catalyzes the synthesis of long-chain saturated fatty acids and is overexpressed during prostatic tumorigenesis, where it therapeutic target in several ongoing trials. However, mechanism FASN upregulation prostate cancer remains unclear. Here, we examine FASN gene CpG methylation pattern by InfiniumEPIC profiling whole-genome bisulfite sequencing across multiple racially diverse primary metastatic cohorts, comparing with protein expression as measured digitally quantified IHC assay reverse phase array analysis or expression. We demonstrate that body hypomethylated tumors compared benign tissue, significantly inversely correlated both cancer. Primary ERG rearrangement have increased find evidence hypomethylation this context. also from carriers germline HOXB13 G84E mutation matched controls, consistent a report HOXB13 may contribute to epigenetic regulation in vitro. contrast previous studies, no significant association self-identified race models include status two independent tumor cohorts. Taken together, these data support potential for which be relevant selecting patients responsive inhibitors.
Significance:
Here, leverage cohorts highly This finding shed light on mechanisms provides potentially useful biomarker future trials inhibitors.
