ABSTRACT
This multicenter, dose-ranging study evaluated the antihypertensive effectiveness of once-daily administration fosinopril sodium in 220 patients with supine diastolic blood pressure 95-115 mm Hg. After a 4-week placebo period, were randomly assigned to double-blind therapy either or 10, 40, 80 mg once daily for 4 weeks. If treatment goals not met, chlorthalidone 25 mg/day was added weeks 5 8. Thereafter, could enter long-term, open-label phase and receive 10-80 plus chlorthalidone, if needed. monotherapy, average decreases 9% (10 mg), 11.5% (40 12.5% (80 mg) compared 6% group. 8 weeks, decreases, without diuretic therapy, 12.5-18.2%, 10.8% placebo. Blood continued be well controlled, showed no evidence tachyphylaxis tolerance through 12-15 months treatment. Fosinopril tolerated. During short-term phase, patient withdrew because adverse events possibly related fosinopril; during long-term nine 148 (6.1%) that reason. In mild-to-moderate hypertension, provided significant effects therapy. The 10 dose effective some may considered starting dose.
