ABSTRACT
Clinical states in which angiotensin II is increased are often associated with increases mineralocorticoids. To determine the effects of mineralocorticoids on action, we examined aldosterone receptor expression and function cultured rat vascular smooth muscle cells. Incubation resulted concentration- time-dependent number, without changes binding affinity. For example, incubation 1 microM for 40 hours 59% number. Increases receptors were dependent protein synthesis as evidenced by time dependency upregulation inhibition cycloheximide. enhanced II-stimulated phospholipase C activation, demonstrated II-induced inositol phosphate responses proportion to In addition, prevented downregulation surface desensitization formation. summary, 1) directly 2) responses, 3) desensitization. conclusion, may potentiate pressor via receptors.
