ABSTRACT
The purpose of this study was to measure the temporal profile expression endothelial nitric oxide synthase (NOS) in cerebral microvessels after middle artery occlusion rat. Middle performed on 24 male Wistar rats by extracranial insertion a 4-0 nylon monofilament into internal artery. Three additional were used as controls. Animals killed at 1, 2, 4, 6, 24, 48, 72, and 168 hours (n = 3 per time point). Rat brains perfused with buffer, frozen, sectioned, stained monoclonal antibody against NOS. Adjacent sections hematoxylin eosin for evaluation neuronal damage. NOS vessels upregulated 1 hour induction ischemia throughout ischemic region. progressively increased up ischemia. In periphery area necrosis cortex, delayed (24-hour) upregulation remained constant duration rapid intense differential core peripheral areas lesion indicates role cell damage suggests that may mediate changes blood flow.
