ABSTRACT
The specificity of T cell help for B activation and differentiation is maintained by the brief expression on surface, following receptor-mediated triggering, CD40 ligand (CD40L). Interaction helper (Th) CD40L with induces activation, surface antigens, proliferation, initiation immunoglobulin isotype switch. We predicted that in patients systemic lupus erythematosus (SLE), whom Th cell-dependent production autoantibodies results immune complex-mediated tissue damage, might be augmented, prolonged, or abnormally regulated. Baseline was increased some SLE studied, when compared control subjects. While cells from normal subjects (n = 14) rheumatic disease 9) showed maximal CD40L, after vitro phorbol myristate acetate (PMA) ionomycin, at 6 h culture diminished levels observed 24 48 h, 19) high level through culture. prolonged functionally significant, as h-activated cells, cocultured target induced greater CD80 (B7-1) than did cells. These document impaired regulation identify an important potential therapy this autoimmune disease.
