MiR-613 inhibits proliferation and invasion and induces apoptosis of rheumatoid arthritis synovial fibroblasts by direct down-regulation of DKK1

ABSTRACT
This study aimed to investigate the effects of miR-613 on proliferation, invasion and apoptosis rheumatoid arthritis synovial fibroblasts (RASFs).Synovial tissue samples were collected from 20 (RA) patients 10 with joint trauma undergoing replacement surgery. The RASFs isolated cultured. MiR-613 DKK1 expression in both tissues cells was detected using quantitative real-time PCR (qRT-PCR). Dual luciferase reporter gene assay employed evaluate effect activity DKK1. Then transfected mimics, si-DKK1 pcDNA-DKK1. Changes cellular through BrdU assay, Transwell flow cytometry analysis, respectively.MiR-613 significantly down-regulated RA compared normal cells, whereas up-regulated RASFs. showed that could specifically bind 3'UTR inhibit activity. Moreover, reduced Overexpression or knockdown suppressed proliferation RASFs, induced RASF apoptosis. reverse results observed when miR-613-overexpressing RASFs.MiR-613 can induce by directly targeting expression.
