ABSTRACT
Diazotrophic (nitrogen-fixing) Gram-positive and endospore-formed Paenibacillus spp. have potential uses as a bacterial fertilizer in agriculture. The transcriptional analysis of nitrogen fixation is lacking, although regulation mechanisms been well studied Gram-negative diazotrophs.Here we report global profiling sp. WLY78 cultured under N2-fixing condition (without O2 NH4(+)) non-N2-fixing (air 100 mM NH4(+)). nif (nitrogen fixation) gene operon composed 9 genes (nifBHDKENXhesAnifV) this bacterium was significantly up-regulated compared to condition, indicating that transcription strictly controlled by NH4(+) O2. qRT-PCR confirmed these were differently expressed. Non-nif specifically required fixation, such mod, feoAB cys encoding transporters Mo, Fe S atoms, coordinately transcribed with condition. transcript abundance suf specific for synthesis Fe-S cluster suggesting Sul system, which takes place nifS nifU, plays important role the nitrogenase.
