ABSTRACT
Abstract Background Liquidambaris Fructus (LF) is the infructescence of Liquidambar formosana . In Traditional Chinese Medicine, LF has been used to treat joint pain, a common symptom arthritis and rheumatism; however, lack pharmacological evidence limited its applications in modern clinics. Therefore, this study aims explore protective effect on rheumatoid (RA) identify active ingredients. Methods Rats with adjuvant-induced (AIA) were divided into 4 groups administered petroleum ether extract (PEL), ethyl acetate (EEL), water (WEL), or piroxicam (PIR) respectively for 3 weeks. Two additional as normal control (NC) model (MC) distilled placebo. The clinical scores arthritis, bone surface, synovial inflammation cartilage erosion evaluate therapeutic efficacy each treatment. serum IL-1β TNF-α level expression NLRP3, caspase-1 p20 tissue AIA rats evaluated by ELISA Western blot. ingredients investigated using network pharmacology molecular docking methods, their inhibition NLRP3 inflammasome activation was verified human fibroblast-like cells (RA-FLS) model. Results PEL could alleviate paw swelling, destruction, rats, significantly superior that EEL WEL. reduced levels, attenuated upregulation rats. Network results indicated myrtenal β-caryophyllene oxide main two PEL, these compounds showed significant TNF-α, RA-FLS. Conclusions Myrtenal screened from suppress inflammasome, thereby alleviating RA symptoms.
