ABSTRACT
Adiponectin has multiple functions including insulin sensitization, anti-inflammation and antiatherogenesis in various organs. activates 5'-adenosine monophosphate-activated protein kinase (AMPK) peroxisome proliferator-activated receptor (PPAR)α via the adiponectin (AdipoR) 1 2, which are critical for regulating lipids glucose homeostasis controlling oxidative stress. We investigated whether resveratrol can inhibit renal damage type 2 diabetic db/db mice underlying mechanisms of its effects.Four groups male C57 BLKS/J db/m human glomerular endothelial cells (HGECs) were used. Resveratrol was administered to nondiabetic by oral gavage 12 weeks starting at 8 age.In mice, increased serum levels decreased albuminuria, matrix expansion, inflammation apoptosis glomerulus. phosphorylation AMPK silent information regulator T1 (SIRT1), downstream effectors class O forkhead box (FoxO)1 FoxO3a increasing AdipoR1 AdipoR2 cortex. Furthermore, expression PPARγ coactivator (PGC)-1α, estrogen-related receptor-1α, phosphorylated acetyl-CoA carboxylase sterol regulatory element-binding 1. This effect lowered content nonesterified fatty acid triacylglycerol kidneys, decreasing apoptosis, stress activating nitric oxide synthase. prevented cultured HGECs from undergoing high-glucose-induced AMPK-SIRT1-PGC-1α axis PPARα through increases expression.These results suggest that prevents nephropathy ameliorating lipotoxicity, stress, dysfunction expression.
