ABSTRACT
Abstract Background The intricate landscape of immunocytes in the tumor microenvironment (TME) is fundamental to immunotherapy but notably under-researched extrahepatic cholangiocarcinoma (ECCA). Methods Single-cell RNA sequencing technology was conducted make an in-depth analysis from matched tissues, paratumor tissues and peripheral blood ECCA patients. potential cellular interactions between two cell populations were analyzed with software CellPhoneDB (v2.1.7). Results We obtained 13526 cells characterized transcriptomes heterogeneity different clusters subclusters ECCA, including CD4+ T cells, CD8+ B myeloid immunocytes. observed rarely described immunocyte "intermediate" exhausted (CD8+ Tex) “nonclassic” plasmacytes (CD27+ CD138+ CD38−). In addition, we identified targets, for example, ACP5, MAGEH1, TNFRSF9 CCR8 Tregs MT1 Tex cells. also found strong among Treg M2 macrophages through ligand–receptor analysis, implying that cross-linkage promoted immunosuppressive nature TME. Conclusions a word, our study illuminated components TME revealed at individual level aimed provide new perspective further immunological studies ECCA.
