CircASH2L facilitates tumor-like biologic behaviours and inflammation of fibroblast-like synoviocytes via miR-129-5p/HIPK2 axis in rheumatoid arthritis

ABSTRACT
Previous study showed that circular RNA Absent-Small-Homeotic-2--Like protein (circASH2L) was higher in rheumatoid arthritis (RA) patients. However, the roles and mechanisms of circASH2L RA progression remain unclear.Levels analysis conducted using western blot qRT-PCR. The proliferation, apoptosis, cell cycle progression, migration, invasiveness, inflammation fibroblast-like synoviocytes (RA-FLSs) were determined via MTT, flow cytometry, blot, transwell, ELISA assays.CircASH2L knockdown RA-FLSs suppressed proliferative, migratory, invasive capacities, triggered arrest, promoted inhibited inflammation. Mechanistically, targeted miR-129-5p, repression miR-129-5p abolished functions silencing on growth, motility, RA-FLSs. Besides, found to directly target HIPK2, tumor-like biologic behaviors regulating HIPK2. Importantly, we proved could modulate HIPK2 expression miR-129-5p.CircASH2L RA-FLS through miR-129-5p/HIPK2 axis.
