ABSTRACT
Abstract Background Polycystic ovarian syndrome (PCOS) accounts for about 75% of anovulatory infertility. The cause PCOS is not clear. CircRNAs acting as miRNA sponges mediate the post-transcriptional regulation multiple genes. CYP19A1 a limiting enzyme in steroidogenesis pathway. However, mechanism circRNAs regulating granulosa cell (GC) estradiol secretion remains to be elucidated. Methods Bioinformatics was used predict potential target miRNAs circ_0043532 and genes miR-1270. Target mRNA expression were verified by qRT-PCR GCs from 45 women with 65 non-PCOS. Western blot, ELISA dual-luciferase reporter assays applied confirm substrate Results Circ_0043532 significant up-regulation patients PCOS. predicted circ_0053432, miR-1270, miR-576-5p, miR-421 miR-142-5p, notably decreased Mechanistic experiments showed that specifically binds MiR-1270 negatively regulated circ_0043532. Concomitantly, miR-1270 inhibited production, which could reversed over-expression. Conclusion We identified circ_0043532/miR-1270/CYP19A1 axis contributes aberrant This study broadens spectrum pathogenic factors PCOS, might therapeutic
