ABSTRACT
Antimicrobial peptides (AMPs) are instrumental in maintaining intestinal homeostasis and have emerged as potential therapeutic candidates for ameliorating bacterial infections. However, the intrinsic instability associated with vivo delivery of AMPs constitutes a substantial impediment to their efficacy treating In this study, we genetically modified Bacillus subtilis (B. subtilis) WB600 express Zophobas atratus defensin (ZD), an antimicrobial peptide broad-spectrum activity isolated from atratus, oral administration. This engineered strain effectively protects against Salmonella Infantis (S. Infantis) infection mice. Pretreatment WB600/ZD prevented NF-κB pathway activation induced by S. increased expression antioxidant tight junction proteins, thus alleviating severity inflammation both jejunum ileum (P < 0.01). Moreover, pretreatment facilitated growth beneficial bacteria such Lachnospiraceae, Butyricicoccus, Eubacterium_xylanophilum, Clostridia_UCG-014 while decreasing abundance pathogenic Escherichia-Shigella 0.05). conclusion, study underscores protective effects on Infantis-induced inflammation, suggesting that B. may be promising prophylactic strategy combating infections intestine.
