ABSTRACT
Abstract Background Nematode egg parasitic fungus, Purpureocillium lilacinum is the most effective biocontrol agent and has been widely used commercially in many countries for management of root-knot nematode, Meloidogyne incognita . Availability indigenous potential strains specific to an agro ecosystem very crucial their successful commercial exploitation suppression nematode population. Hence, attempt was made isolate, characterize, evaluate identify P. from pulse chickpea. Results The fungal colony initially white when spore formed it turned into pink colour 72 96 h. Hyphae hyaline septate, conidiophore blunt, phialides were with wide base long neck bearing round oval conidia chains. Molecular identification species, carried out based on ITS1-5.8S-ITS2 region genomic DNA. In vitro bioassay cultural filtrates juvenile mortality revealed that maximum percentage observed IIPR-Pl-11 (88.36%). Spectrophotometric assay chitinolytic activity showed strain produced significantly high activity, chitinase enzyme total protein content (0.139, 51.1 and173.75 µg/ml at 5days); (0.245, 90.1 272.67 10 days) (0.273, 100.4 306.25 µg/ml), respectively, 15 days culturing colloidal chitin-enriched medium C. 2D gel electrophoresis crude suspension presence (32, 46 kDa size) sample chitinase-induced fungus grown C inhibition hatching highest percent by IIPR-Pl-8 5 inoculation (42.6%) 20 (62.80 93.50%), respectively. vivo pot experiment among all strains, efficient promoting plant growth effectively reducing gall number (41.3 per plant), mass (28.3 plant) soil population (284.3 200cc soil) compared control. Conclusion rhizosphere M.
