ABSTRACT
Ceramides (CERs) play key roles in signal transduction and cell regulation, probably during the keratinization of human hair. Current methods using mass spectrometry (MS), however, are not sufficient to allow comprehensive analysis CER molecules, including isobaric isomeric CERs. Therefore, a method for profiling CERs was developed. The developed is based on reversed-phase liquid chromatography (RPLC) coupled atmospheric pressure chemical ionization (APCI)-MS. Comprehensive identification achieved two sets multimass chromatograms obtained from channel detections that monitor both molecular-related sphingoid-related ions under different in-source collision-induced dissociation conditions retention times RPLC. application this revealed hair contains 73 species which were all corroborated by structural tandem spectrometry. results further composition characterized predominant molecules consisting even carbon atom-containing saturated/unsaturated nonhydroxy or α-hydroxy fatty acids C18 dihydrosphingosine, minor but distinct content isobaric/isomeric odd chain-containing This successfully RPLC-APCI-MS technique allows investigation their physicochemical physiological roles. (CERs), consist long amino alcoholic chain [sphingoid moiety (SPM)] covalently bound via an amide linkage acid (FAM), have important barrier function water-holding property skin, together with other lipids intercellular spaces stratum corneum (1Elias P.M.
