ABSTRACT
A NMR spectroscopic method is described that enables the quantitation of specific lipid classes and components, independent fatty acid composition. We demonstrate this for measuring cholesterol, squalene, pools sterol esters, wax esters (WEs), triglyceride (TG) components in sebum meibum. When 600 MHz equipment used conjunction with highly sensitive cryogenically cooled probes, has adequate sensitivity, some applications, advantages over commonly HPLC-evaporative light-scattering detection mass spectrometry-based approaches. This shown to be useful preclinical clinical monitoring efficacy sebum-reducing agents animals humans. In Syrian hamsters, 3% topical flutamide 20 mg/kg oral isotretinoin reduced by 18.7% 30.0%, respectively, WEs 32.9% 31.8%, as measured a punch biopsy ear. 72 patient methodology study, assay delivered reproducible noninvasive measurements WEs, cholesteryl TGs, squalene from Sebutape skin blots. The quantitative results analysis obtained correlate well those HPLC-based approach may broadly applicable cases which acid-independent quantification desired.
