Prognostic importance of cell-cycle activity and genotype in gastrointestinal stromal tumors.

ABSTRACT
e20501 Background: Size, mitosis and locus are important for risk stratification of gastrointestinal stromal tumors (GIST). Genotype cell cycle suggested related to aggressiveness progression, considered have prognostic impact, although most reports analyzed expression proteins or mRNAs. In this study, we examined the biological values CDK activities in primary GIST with R0 surgery. Methods: Histologic Dx was done HE IH. Using lysate frozen materials from 76 pts (n=70), leiomyoma (2), schwannoma (3) abdominal sarcoma (1), CDK-specific were measured by C2P system (activities CDK1 & CDK2). The median age 64 yrs 33 (45%) male. follow-up 37 months. Results: KIT PDGFRA mutations found 57 7 pts, respectively, deletion codon 557-558 16 pts. Two pediatric GISTs one NF-1 had no mutation both genes. measurable 63 (86%) CDK2 (19 U 81) not different those other (18 47). GIST, 20 recurrences 6 deaths observed. Size (p=0.0001), location (p=0.0007), (p=0.0044), rupture (p=0.0105), (p=0.0037), (p=0.0005) ratio (CDK2/CDK1) (p=0.0004) significantly recurrence. Cox regression analysis, only (p=0.0189) (p=0.0070) univariant correlated size, mitosis, Ki-67, location, rupture, and, multiple (p=0.0384), marginally (p=0.0806). mutations, significant factors cause-specific survival. Conclusions: activity predictive recurrence after surgery GIST. Genotyping analysis may be stratification.
