MiRNAs hsa-miR-30a-5p and hsa-miR-99b-5p regulate chemokine-mediated immune response in colorectal cancer.

ABSTRACT
e15500 Background: The immune response plays a key role in the oncogenesis of colorectal cancer. heterogeneity tumor environment determines need for immunogenetic profiling to identify associations with implementation tumor-specific responses purpose our study was describe expression profiles miRNAs and genes involved regulation Methods: included 18 patients (median age 66 years) diagnosed cancer who were treated at National Medical Research Centre Oncology 2018-2019. Total RNA isolated using TRIzol (Thermo Fisher, USA) followed by treatment DNase 1 (ThermoFisher, USA). Sequencing samples performed two approaches: AmpliSeq Illumina Immune Response Panel (Illumina, TruSeq Small Library Prep Kit -Set A according manufacturer's instructions on NextSeq 550 device (llumina, Results: revealed 168 differentially expressed 46 (p < 0.05). 29 miRNA-mRNA pairs identified. It has been established that signaling mechanism is chemokine pathway (hsa04062). Expression CXCL1 CXCL10 encoding chemokines same name increased cells (logFC = 1.51, p 0.006 logFC 2.67, 0.001, respectively). At time, level hsa-miR-30a-5p hsa-miR-99b-5p, which negatively regulate CXCL10, decreased -2.26, 0.001 -1.57, 0.001). Conclusions: Activation simultaneous loss negative regulators hsa-miR-99b-5p determined tumors NGS-sequencing, seems promising development personalized therapy, based features colon tumors.
