Influence of APOE genotype on familial aggregation of AD in an urban population

ABSTRACT
<b><i>Objective:</i></b> To examine the influence of proband's <i>APOE</i> genotype on AD among first-degree relatives in a community-based study African Americans, whites, and Caribbean Hispanics. <b><i>Methods:</i></b> History demographic information were obtained 1,073 siblings parents 312 patients with 2,722 802 nondemented controls. genotyping was performed all 1,114 <b><i>Results:</i></b> A higher proportion (35%) than controls (27%) had one or more<i> APOE</i>-ε4 alleles (<i>p</i> = 0.03). When compared without an<i> allele, risk for increased both (rate ratio [RR] 1.9, 95% confidence interval [CI] 1.2 to 3.1) (RR 1.8, CI 2.6) more <i>APOE</i>-εe alleles, regardless ethnic group. There similar trend an <i>APOE</i>-ε4 but this limited Hispanics Americans. <b><i>Conclusions:</i></b> The presence allele increases relatives, probands' disease status, groups. Relatives also at suggesting that other genes factors may risk.
