ABSTRACT
Asymmetric expression of sonic hedgehog (Shh) in the left side Hensen's node, a crucial step for specifying left-right (LR) axis chick embryo, is established by repression Shhexpression right node. The transcriptional regulator that mediates this has not been identified. We report isolation and characterization novel Polycomblike 2 gene, Pcl2, which encodes transcription repressor displays an asymmetric expression, downstream from Activin-βB Bmp4, node developing embryo. In vitro mapping studies define activity to PHD finger domain Pcl2 protein. Repression Pcl2expression early embryo results randomized heart looping direction,which accompanied ectopic Shh genes lateral plate mesoderm (LPM), while overexpression represses was also supported overexpressed limb bud feather bud. Similarly,transgenic mouse inhibits ZPA. pull-down assays demonstrated direct interaction with EZH2, component ESC/E(Z) repressive complex. Taken together fact found directly repress promoter vitro, our demonstrate role regulating LR patterning silencing
